How would you compare an S40 vs a Triathlon Bike?

All things the same from a hardware perspective, same person, power output, etc. I know the V20 performs really well against these bikes, but how much does the 20 vs 40 angle difference matter for overall speeds? I know the S40 is built more for touring, but I still think it should hold well against these bikes.

For reference, the ad on the front page shows a V20 and a triathlon bike, so it made think about an S40 and tri bike matchup.

Sorry but the question is too vague to bear any meaning. What do you want to do with your bike? Ride a triathlon? Which format? On the flat or rather hilly?
In most cases, the best bike will be the one one which you’ve spent most time training.

I would say the S40 can hold its own against many regular riders on a ti-bike given the same fitness, etc. Not V20c fast but up there. It would be unfair to compare a very very good upright rider to an S40 - mostly as those pros and semi-pro try riders basically live on their road bikes and are hyper-trained to that one thing, so...

The X-factor comes in comfort wise - and I can ride the S40 all day and definitely much longer without discomfort than an upright.

I haven't seen any CdA data from S40 riders, but IIRC some of the lowest CdAs that I have seen from TT/Tri riders are barely under 0.170 in a wind tunnel, and again, IIRC Larry and Maria were close to or maybe even just under 0.160 on their V20s.

I believe I am taller and heavier, and other than my data coming from MyWindSock on a course with variable wind directions on an up and down course (instead of a velodrome or a wind tunnel) my slipperiest ranges from 0.180-0.190.

Now, if someone with an S40 with decently fast wheels and a helmet, and the My Windsock app can either get on a velodrome, something similar or do a few runs back to back up the same course then we can at least get a starting point to go from. But yeah, I think the S40 would do very well against a TT/Tri bike. A bonus it that you don't have to wear one of those huge round mailboxes on your head either.
